Description

Description: This assay employs a two-site sandwich ELISA to quantitate HIST1H2AD in samples. An antibody specific for HIST1H2AD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any HIST1H2AD present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HIST1H2AD is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HIST1H2AD bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. This structure consists of approximately 146 bp of DNA wrapped around a nucleosome, an octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures.
HIST1H2AD is intronless and encodes a member of the histone H2A family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element. This gene is found in the large histone gene cluster on chromosome 6p22-p21.3
